pCCI-4K-SARS-CoV-2-Wuhan-Hu-1 (GenBank accession MT926410) is a plasmid containing a cDNA copy of the full-length genome of SARS-CoV-2-Wuhan-Hu-1, flanked by a CMV promoter and a hepatitis delta virus ribozyme to drive the production of authentic full-length viral RNA in transiently transfected cells. The system facilitates reliable rescue of SARS-CoV-2 from a single plasmid (no in vitro transcription or helper plasmids required) and is easy to manipulate for reverse genetics studies.

Reporter virus derivatives containing the mCherry (pCCI-4K-SARS-CoV-2-mCherry, GenBank accession MT926411) or NanoLuc luciferase (pCCI-4K-SARS-CoV-2-NLuc, GenBank accession MT926412) reporter inserted between the ORF7a and ORF7b genes are also available.

The pCCI-4K system is designed for propagation in EPI300 E. coli (not included) facilitating genetic manipulation at ~1 plasmid copy/cell whilst allowing higher copy propagation (for DNA extraction), when cultured in the presence of arabinose. The pCCI-4k plasmids contain a chloramphenicol selection marker.
